A PhD Fellowship on “Improving CRISPR genome editing efficiency by modifying epigenetic marks” in the Brakebusch Group at BRIC, University of Copenhagen

The group and our research 
The Brakebusch group (www.bric.ku.dk/Research/Brakebusch_Group/) is using CRISPR technology to make genetically modified mice, primary cells and cell lines. Cord Brakebusch is also head of the Transgenic Mouse Core Facility at the University of Copenhagen (www.transgenicmice.ku.dk) where technology development projects are exploring the generation of mice with multiple fluorescent reporters and of ES cells with multiple disease-related mutations. 
 
Project/ The research project(s) 
Epigenetic modifications of DNA and histones are likely to influence the efficiency of CRISPR genome editing by homologous directed recombination or non-homologous end joining (NHEJ). Furthermore, epigenetic changes triggered by genome editing might influence the expression of the recombined gene. To investigate the relationship between epigenetic marks and CRISPR genome editing efficiency, we will compare global and target gene specific epigenetic modifications in cell lines with high and low CRISPR genome editing efficiency. We will then try to improve genome editing efficiency by manipulating epigenetic marks on a global and on a target gene specific level. For the latter task, cutting-deficient dCas9 coupled to enzymes altering epigenetic marks will be used. We expect to obtain novel insight into molecular mechanisms regulating CRISPR genome editing efficiency and to find conditions to improve precise genome editing.

Eligibility criteria 
 
It is a prerequisite for applying for the PhD-fellowship that the following conditions are fully met: 
 
1. Early-stage researchers (ESR) are those who, at the time of recruitment by the University of Copenhagen, are in the first four years (full-time equivalent) of their research careers. This is measured from the date when they obtained the degree which formally entitles them to embark on a doctorate, either in the country in which the degree was obtained or in the country in which the research training is provided, irrespective of whether or not a doctorate was envisaged. Please note that applicants cannot already hold a PhD at the time of recruitment 
2. Conditions of international mobility of researchers: The early-stage researchers are required to undertake transnational mobility (i.e. move from one country to another) when taking up the appointment.  ESRs must not have resided or carried out their main activity (work, studies, etc.) in the country of their host organisation for more than 12 months in the 3 years immediately prior to their recruitment. Short stays, such as holidays, are not taken into account. 
 
Qualifications  
We are looking for a highly motivated and enthusiastic candidate with the following qualifications: 
A Master of Science in biology, biochemistry, molecular medicine, or similar 
Experience in molecular and cellular biology 
Excellent communication skills in English

Place of employment and work  
The employment and main place of work is at BRIC, University of Copenhagen. BRIC is located close to the centre of Copenhagen, in the Copenhagen Biocenter. Secondments at other institutions within the IMGENE consortium are a mandatory part of the successful candidate's employment. Accordingly, the selected PhD candidate is expected to spend several month studying and performing interdisciplinary research at different IMGENE institutions.

Terms of salary and employment 
The terms of employment and salary are in accordance with the agreement between the Ministry of Finance and The Danish Confederation of Professional Associations on Academics in the State and according to the rules and regulations laid down by European Union’s Horizon 2020 Marie Skłodowska-Curie Initial Training Networks. 
The appointment as PhD fellow is for 3 years. It is conditioned upon the candidates’ success­ful enrolment in the PhD programme. This requires submission and acceptance of an application for the specific project formulated by the candidate. The PhD study must be completed in accordance with The Ministerial Order on the PhD programme (2013) and the University’s rules on achieving the degree.

The further process 
Shortlist: After expiry of the application deadline, the superior with power to appoint selects a number of applicants for assessment on the advice of the Appointments Committee. All applicants are immediately notified whether their application has been passed for assessment. Applicants who were not passed for assessment should not expect further with regard to their application unless the shortlist is revised. 
 
Assessment: The selected applications will be assessed according to the Ministry Order on the Appointment of Academic Staff at Universities 2012 and the University of Copenhagen’s guidelines 2013. The Assessment Committee makes a non-prioritized assessment of the academic qualifications and experience with respect to the above mentioned area of research, techniques, skills and other requirements listed in the advertisement.
